miR-339-3p inhibits proliferation and metastasis of colorectal cancer

چکیده
MicroRNAs (miRNAs) serve important roles in regulating cancer cell proliferation and metastasis. The same hairpin RNA structure may produce mature products from each strand, termed miR-5p and miR-3p, which can bind different mRNAs. Previously, the present authors reported that miR-339-5p could inhibit cell proliferation and migration by targeting the 3'-untranslated region (3'-UTR) of PRL-1 mRNA. The present study analyzed the expression, function and preliminary regulatory mechanism of miR-339-3p in colorectal cancer (CRC). The results of reverse transcription-quantitative polymerase chain reaction analysis demonstrated that miR-339-3p is downregulated in CRC specimens and highly invasive cell lines. Furthermore, the low-level expression of miR-339-3p was significantly associated with lymph node metastasis in patients with CRC; however, reduced miR-339-3p expression was not associated with age, gender or the differentiation status of the tumor. Overexpression of miR-339-3p was sufficient to suppress tumor growth and metastasis in vitro. In addition, the present study demonstrated that unlike miR-339-5p, PRL-1 expression was not regulated by miR-339-3p. The findings of the present study indicate that miR-339-5p and miR-339-3p may target different mRNA. The target gene of miR-339-3p requires future identification.

miR-506 inhibits cell proliferation and invasion by targeting TET family in colorectal cancer
Objective(s): Ten-eleven translocation (TET) family members have been shown to be involved in the development of many tumors. However, the biological role of the TET family and its mechanism of action in colorectal carcinogenesis and progression remain poorly understood. Materials and Methods:We measured the expression levels of TET family members in colorectal cancer (CRC) specimens, in the c...
متن کاملMiR-590-3p promotes proliferation and metastasis of colorectal cancer via Hippo pathway
Studies reported that miR-590-3p was involved in human cancer progression. However, its roles of oncogene or anti-oncogene in malignancies still remain elusive. This study was aimed to investigate the effect of miR-590-3p on the cell proliferation and metastasis via Hippo pathway in colorectal cancer (CRC). In our study, miR-590-3p was demonstrated highly expressed in CRC tissues, compared with...
متن کاملmicroRNA-202-3p inhibits cell proliferation by targeting ADP-ribosylation factor-like 5A in human colorectal carcinoma.
PURPOSE MicroRNAs (miRNA) that are strongly implicated in carcinogenesis have recently reshaped our understanding of the role of non-protein-coding RNAs. Here, we focused on the function and molecular mechanism of miR-202-3p and its potential clinical application in colorectal cancer. EXPERIMENTAL DESIGN miR-202-3p expression was determined by quantitative reverse transcriptase PCR (qRT-PCR) ...
متن کاملMiR-331-3p inhibits proliferation and promotes apoptosis by targeting HER2 through the PI3K/Akt and ERK1/2 pathways in colorectal cancer.
MicroRNAs (miRNAs) regulate cell proliferation, apoptosis and carcinogenesis by targeting related mRNAs in different types of cancer. miR-331-3p has been found to regulate the development and progression of various types of cancer cells. However, little research has been conducted on the role of miR-331-3p in colorectal cancer (CRC). The present study aimed to explore the function of miR-331-3p...
متن کاملmiR-27a-3p targeting RXRα promotes colorectal cancer progression by activating Wnt/β-catenin pathway
This study aimed to elucidate how miR-27a-3p modulates the Wnt/β-catenin signaling pathway to promote colorectal cancer (CRC) progression. Our results showed that the expression of miR-27a-3p was up-regulated in CRC and closely associated with histological differentiation, clinical stage, distant metastasis and CRC patients' survival. miR-27a-3p mimic suppressed apoptosis and promoted prolifera...
